Methods to analyze extracellular vesicles at single particle level

چکیده

Abstract Extracellular vesicles (EVs) are nano-sized derived from cells that transport biomaterials between through biofluids. Due to their biological role and components, they considered as potential drug carriers for diagnostic applications. Today's advanced nanotechnology enables single-particle-level analysis was difficult in the past due its small size below diffraction limit. Single EV reveals heterogeneity of EVs, which could not be discovered by various ensemble methods. Understanding characteristics single EVs more pathological researches. This review focuses on techniques employed at particle level describes principles each technique.

Methods to isolate extracellular vesicles for diagnosis

Extracellular vesicles (EVs) are small membrane-bound bodies that are released into extracellular space by diverse cells, and are found in body fluids like blood, urine and saliva. EVs contain RNA, DNA and proteins, which can be biomarkers for diagnosis. EVs can be obtained by minimally-invasive biopsy, so they are useful in disease diagnosis. High yield and purity contribute to precise diagnos...

Alternative Methods for Characterization of Extracellular Vesicles

Extracellular vesicles (ECVs) are nano-sized vesicles released by all cells in vitro as well as in vivo. Their role has been implicated mainly in cell-cell communication, but also in disease biomarkers and more recently in gene delivery. They represent a snapshot of the cell status at the moment of release and carry bioreactive macromolecules such as nucleic acids, proteins, and lipids. A major...

Flow Virometry to Analyze Antigenic Spectra of Virions and Extracellular Vesicles

Cells release small extracellular vesicles (EVs) into the surrounding media. Upon virus infection cells also release virions that have the same size of some of the EVs. Both virions and EVs carry proteins of the cells that generated them and are antigenically heterogeneous. In spite of their diversity, both viruses and EVs were characterized predominantly by bulk analysis. Here, we describe an ...

Extracellular Vesicles Derived from Gastrointestinal Microbiota: A New Approach to Clinical Studies

Extracellular vesicles, naturally released from all cell types including bacteria, are of great importance in medical microbiology due to transporting a variety of biomaterials, enzymes, and virulence factors, regulating immunity, and having roles in colonization and initiation of signaling pathways. These vesicles are also secreted from microbiota in the gastrointestinal tract and affect the h...

Extracellular Single-Unit Recording Methods*

Since their refinement in the early 195Os, extracellular, singleunit recording methods have been used to obtain a wealth of data about the properties of CNS structures. The applications of the technique have been diverse: extracellular microelectrodes have been used to map the potential fields of single discharging neurons in order to answer fundamental questions about the excitability of CNS d...
